Abstract

KTIVITAS ANTIOKSIDAN EKSTRAK DAUN KETAPANG (Terminalia cattapa L.) DENGAN BERBAGAI METODE DAN UJI SITOTOKSIKNYA Novel Suci Nurania) , Leny Heliawati a), Siti Warnasih a*) a) Department of Chemistry, Pakuan University, Bogor *) Corresponding Author: sitiwarnasih@unpak.ac.id ABSTRACT Ketapang leaves (Terminalia catappa L.) are wild plants that contain secondary metabolite compounds, namely flavonoids, triterpenoids, and tannins that have potential as antioxidants. Flavonoid metabolite compounds in ketapang leaves are thought to have anticancer activity and reduce cell viability. This study aims to determine the antioxidant and cytotoxic activity of methanol extracts of ketapang leaves (Terminalia catappa L.) and identify secondary metabolite compounds using GC-MS, which later can have potential as anticancer. The study began with maceration extraction using 96% methanol then fractionated with n-Hexan and Ethyl Acetate. Antioxidant activity was tested using DPPH, CUPRAC and FRAP methods. Furthermore, the best antioxidant activity value from the three methods was then tested for cytotoxicity using the BSLT (Brine Shrimp Lethality Test) method and identified active compounds using GC-MS. The results showed that methanol extract, N-Hexan fraction and Ethyl acetate fraction of ketapang leaves gave the highest antioxidant activity using DPPH, CUPRAC and FRAP methods with the best results with levels of 18.2855; 302.7199; and 180.4672 µg vitamin C/mg extract in methanol extract. The results of probit analysis in the cytotoxic test showed that the methanol extract had an LC50 value of 410.96 ppm so that the methanol extract was toxic. There are 23 compounds based on GC-MS tests on methanol extracts, one of which is epicatechin, Epigallocationchin gallate, and piperidine compounds which have potential as anticancer.
