Stingless bee (Trigona sp) propolis from Bintan, Lampung, and Makassar was obtained by maceration extraction with ethanol solvents. Antioxidant activity was measured by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Total phenolic and flavonoid contents were analyzed by Folin-Ciocalteu colorimetric and aluminum chloride (AlCl3) methods. Identification of secondary metabolite extracts was performed by phytochemical screening indicating the presence of alkaloid, flavonoid and polyphenol showed positive result on propolis ethanol extract from Bintan, Lampung and Makassar, while the saponin compounds showed positive result on ethanol extract propolis from Makassar. The result showed obtained values of 158.81 mgGAE/g and 6.16 mgQE/g from Bintan, 327.86 mgGAE/g and 17.08 mgQE / g from Lampung while from Makassar it was valued at 46 mgGAE/g and 10,64 mgQE/g. Ethanol propolis extract potentially as an antioxidant which is categorized as moderate with an IC50 value of 109.14 ppm from Bintan and 130,328 ppm from Lampung, while from Makassar is categorized as weak with an IC50 value of 173,545 ppm. As many as 21 compounds from Bintan were identified, 33 compounds from Lampung and 36 compounds from Makassar that played an active role as antioxidants, one of which was quercetin, luteolin, genistein, and mangiferin.
